Recovery and Pre-purification of Inulinase from Kluyveromyces Marxianus NRRL Y-7571 in Poly (Ethylene Glycol) / Potassium Phosphate Buffer Aqueous Two-phase Systems

In this work, the recovery and purification of inulinase produced by Kluyveromyces marxianus NRRL Y-7571 in solid state fermentation using in aqueous two-phase systems (ATPS) was studied. The effects of concentration of PEG and potassium phosphate were evaluated using experimental design technique. The SAB extraction yielded a purification factor (PF) up to 3.85-fold, using 18% PEG 4000 and 7% (w/w) of potassium phosphate buffer at pH 7.0. The addition of NaCl to the system caused a decrease in the PF of the enzyme, due to denaturation. A combined purification strategy changing the ionic force of the medium, followed by precipitation with n-propyl alcohol (50% concentration and flow rate of 19.9 mL / min) and SAB extraction yielded a purification factor of 5.45-fold and an enzyme recovery of 86.2%.
